褶纹冠蚌外套膜组织培养及蛋白质电泳特性研究

Studies on The Mantle Tissue and Protein Character of Cristaria plicata Leach

采用褶纹冠蚌的外套膜进行组织块培养 ,将培养后的组织块做切片和涂片观察 ,再用胰蛋白酶消化组织块成单细胞 ,培养后进行细胞计数。同时采用聚丙烯酰胺凝胶 (PAGE)电泳技术 ,比较了褶纹冠蚌的外套膜、珍珠囊、珍珠粉蛋白与经过培养的组织块蛋白的变化。试验结果表明 :经过培养的组织块有旺盛的分泌活动 ,并会出现自溶现象。培养过程中细胞数量的增长呈现明显的规律性 ,产生一个由快速增长到逐渐衰退的变化。外套膜与珍珠囊所含蛋白质基本相同 ,培养后的组织块蛋白质组分减少 。

In this experiment, the mantle tissue were cultured, and it′s construction was observed under microscope. The tissue were dispersed into single cell which can be cultured and counted. The character of proteins from mantle, pearl sac, pearl and cultured tissue were compared using PAGE technique. The results showed that the mantle tissue have the phenomenon of secretion and self lysis. The cells divided rapidly at early stage of culture, slow down at later stage. The proteins from mantle and pearl sac were similar while the protein from cultured tissue is much reduced. The PAGE bands of pearl was less than that of mantle and pearl sac.
