肺生物基质及鼠尾胶对日本血吸虫培养细胞影响的研究 被引量:11

摘要 目的 研究细胞外基质 (ECM)对日本血吸虫培养细胞生存、生长的影响 ,佐证 ECM有利于培养细胞的生存、生长。方法 将虫龄 2 1d的日本血吸虫细胞接种于预先涂有肺生物基质、鼠尾胶的培养瓶和小盖玻片上常规培养 ,分别在光镜及扫描电镜下观察、拍照 ,并运用酶细胞化学方法进行培养细胞的乳酸脱氢酶 (L DH)染色。对照组未涂生物基质。结果 与对照组相比 ,实验组细胞饱满 ,弹性好 ,立体感强 ,存活时间长 ,L DH活性强 ,对高温具一定的耐受性 ;尤其是肺基质中培养的细胞 ,酶活性最强 ,培养 6 0 d的细胞仍观察到具有分裂能力。结论  ECM能改善日本血吸虫培养细胞生存、生长的微环境。实验组间比较 。 Objective To study the effects of extracellular matrix (ECM) on the survival and growth of cultured cells from Schistosoma japonicum(S.japonicum), and to demonstrate that ECM was benefit to the survival and growth of the cultured cells.Methods The cells from 21 day old S. japonicum were inoculated in the flask and on the glass coverslips which was smeared with the bio matrixes from the lung of a rabbit and collagen from rat tail, and then cultured routinely. The cells were observed and photographed by using the microscope (Olympus IM) and scanning electron microscopy (SEM), and stained to show the activities of LDH after 7 days by using cytochemistical method of enzyme. The control group was not smeared with the bio matrixes.Results The cultured cells from S. japonicum in the test group were healthy, solid and good heat resistant, and maintained viability for a longer time. Their LDH activities were strong. Activities of the cells cultured in the lung bio matrix were the strongest, and it was observed that the cells cultured for 60 days were dividing.Conclusions ECM can meliorate micro circumstance for the survival and growth of the cultures from S. japonicum. The matrix from the rabbit lung was a better bio matrix than collagen from rat tail.
