重型肝炎患者肝组织基质金属蛋白酶-1及其抑制因子-1 mRNA的表达

The expression of MMP-1 and TIMP-1 mRNA in patients with viral hepatitis gravis

目的 观察重型肝炎患者基质金属蛋白酶 1(MMP 1)和金属蛋白酶组织抑制因子 1(TIMP 1)mRNA的表达以及胶原蛋白的沉积情况 ,从胶原降解的角度探讨重型肝炎致肝纤维化的机制。方法 重型肝炎肝组织标本 2 0例 ,非肝病患者肝组织 6例 ,用原位杂交法检测肝组织MMP 1和TIMP 1mRNA的表达 ,并作Ⅰ、Ⅳ型胶原的免疫组化以及天狼红胶原染色。结果 与对照组比较 ,重型肝炎组的MMP 1mRNA和TIMP 1mRNA表达显著增加 ,肝组织胶原蛋白总含量和Ⅰ、Ⅳ型胶原蛋白含量亦显著增加。肝组织胶原蛋白总含量和Ⅰ、Ⅳ型胶原蛋白含量与MMP 1mRNA表达无明显相关性 ,与TIMP 1mRNA表达呈正相关。结论 重型肝炎患者肝纤维化的发生与金属蛋白酶抑制因子增加、抑制金属蛋白酶的活性、降低间质胶原蛋白的分解有关。

Objective To study on the possible fibrosis mechanism in the way of degradation of collagen ,the expressions of matrix metalloproteinase 1(MMP 1) mRNA and tissue inhibitor of matrix metalloproteinase 1?mRNA(TIMP 1) and the collagen deposition in the liver tissues.Methods The specimens of the liver tissues in 20 cases with viral hepatitis gravis and 6 cases without liver diseases were detected for the expressions of MMP 1 and TIMP 1 mRNA ,and contents of type I,Ⅳ collagen proteins by in situ hybridization and immunohistochemical staining. Meanwhile the total collagen content was detected by Sirus red staining.Results The total collagen content and quantification of type I, Ⅳcollagen in the patients with hepatitis gravis were significantly higher than those in control group. The expressions of MMP 1 and TIMP 1 mRNA were significantly higher than those in control group. There was a positive correlation between the expression of TIMP 1 and total collagen protein or type I, Ⅳ collagen .The correlation between the expression of MMP 1 mRNA and collagen protein deposition was not found.Conclusion Hepatic fibrosis in the patients with viral hepatitis gravis may be relative to increase of TIMP 1 mRNA expression that inhibit the degradation of collagen.
