摘要
目的 检测铁缺乏大鼠耳蜗肌动蛋白的变化 ,探讨铁缺乏引起耳蜗毛细胞损伤的发生机制。方法 应用免疫组织化学染色法、SDS -PAGE和Westernblot,检测正常及以缺铁饮食饲养 8wk大鼠的耳蜗肌动蛋白水平的变化并进行比较。结果 出现感音神经性耳聋的铁缺乏组大鼠耳蜗肌动蛋白的相对含量降低、免疫组化的反应性明显减弱 ;听力正常的铁缺乏大鼠组中两者无明显变化。结论 耳蜗肌动蛋白含量及免疫组化染色反应性的变化 。
Aim To analyze the change of cochlear actin level in iron deficient rats.Methods Cochlear actin of normal rats and rats fed on iron deficiency diet for 8 weeks was determined by immunohistochemical staining, SDS PAGE and Western blot. The relationship between the quntity and distribution of cochlear actin and the change of auditory brainstem reaction (ABR) threshold was observed. Results 36.1%(13/36 ears)iron deficiency (ID) rats had substantial elevation of ABR hearing threshold. No significant change of cochlear actin expression was observed in iron deficicient rats with normal hearing compared with normal control ( P >0.05), while the quantity of actin dropped obviously in iron deficient rats with hearing loss. The immuno histochemical reaction for actin in cochlear hair cells were also weak in iron deficient rats with hearing loss compared with normal control and iron deficient rats with normal hearing ( P <0.01) . Conclusion The decreased actin in chochlea may be one of important pathological basis for sensorineural hearing loss in iron deficient rats.
