摘要 目的 探讨端粒酶在小儿急性白血病中的表达及作为预测病情、判断预后标志物的可能性。方法 采用端粒重复序列扩增 微孔杂交法 (TRAP Kyb) ,检测 16例急性白血病患儿骨髓和外周血单个核细胞的端粒酶活性。对照组为10例非白血病血液病患儿的骨髓及 5例正常健康小儿的外周血单个核细胞。结果  16例急性白血病患儿表达阳性率83.1% ) ;10例非白血病血液病患儿有 2例呈低度阳性表达 ,5例正常健康小儿外周血单个核细胞中未见阳性表达。结论 端粒酶活性在急性白血病患儿骨髓和外周血单个核细胞中明显升高 ,表明端粒酶的活性与急性白血病的发生可能有着密切关系 。 Aim To investigate the expression of telomerase in acute leukemia of childhood, and the possibility of applying it as a marker for predictating disease progression and prognosis. Methods The telomerase activity in mononuclear cells was determined by using TRAP Kyb in bone marrow from 16 acute leukemia childhood, compared with that in peripheral blood from 10 non leukemia hemopathic childhood and 5 healthy children. Results The positive rate of telomerase activity was 83.1% in 16 acute leukemia childhood; and 2 cases in 10 non leukemia hemopathic childhood were low expression, while telomerase activity was not detected in 5 normal controls. Conclusion Telomerase activity is highly expressed in bone marrow and peripheral blood mononuclear cells from acute leukemia childhood, indicating that telomerase activity close relevance to the pathogenesis of acute leukemia. Detection of telomerase activity in bone marrow may be a applicable marker for evaluating disease progression and prognosis of acute leukemia.
