内皮素-1对大鼠睾丸间质细胞睾酮分泌的可能作用方式 被引量:1

摘要 目的 :了解内皮素 - 1影响离体大鼠睾丸间质细胞睾酮分泌的可能作用方式。方法 :采用大鼠睾丸间质细胞体外孵育技术以及12 5I-睾酮放射免疫法 ,观察了缩血管肽内皮素 - 1 (ET- 1 )对间质细胞睾酮的分泌。结果 :ET- A、ET- B受体拮抗剂均可逆转 ET- 1对离体大鼠间质细胞睾酮分泌的抑制作用 ,但 T-型 Ca+ +通道阻滞剂异搏定 (VER)未表现此作用。结论 :ET- 1通过与 ET- A受体及 ET-B受体结合而发挥作用。其 Ca+ +的动员可能不是通过 T-型 Ca+ +通道的开放。 Objective: To explore the mechanism endothelin-1 affect the testosterone secretion of rat Leydig cells. Methods: After rat Leydig cells were incubated, testosterone was determined by radioimmunoassay. Results: ET-A or ET-B receptor antagonist could reverse the inhibitory effect of ET-1 on testosterone secretion, but Verapamile, a calcium-channel blocker, could not inhibit. Conclusion: ET-1 inhibits testosterone secretion of rat Leydig cell by combining to ET-A or ET-B receptor. Mobilization of Ca ++ may not via opening T- calcium-channel.
