摘要
目的 研究化疗药物 5 - Fu对肝癌细胞内凋亡抑制蛋白 Survivin的诱导作用 ,探讨 Survivin在肝细胞癌化疗抵抗中的作用机制。方法 培养人肝癌细胞株 Hep G2 ,加入低浓度的化疗药物 5 - Fu,分别在加药前、加药后 2 4 h和 4 8h采用逆转录聚合酶链反应 (RT- PCR)和免疫印迹技术 (western- blot)检测 Survivin m RNA和蛋白的表达。结果 给予低浓度的 5 - Fu可诱导 Survivin m RNA和蛋白的表达发生变化 ,其中 m RNA在加药后 2 4 h和 4 8h分别较加药前提高了 1.2倍和 3.9倍 ,蛋白在加药后 2 4 h和 4 8h分别较加药前提高了 0 .9倍和 4 .7倍。结论 5 - Fu可诱导肝癌细胞内 Survivin的表达增加 ,抵抗化疗药物诱导的细胞凋亡。
Objective To investigate the expression of survivin induced by chemotherapy agent(5 Fu),and determine the role in anti chemotherapy of hepatocellular carcinoma(HCC).Methods Low concentration of 5 Fu was added in cultured hepatocarcinoma cell line(HepG 2).The expression of survivin was detected by RP PCR and western blot on 24h and 48h after adding 5 Fu.Results Survivin mRNA level was increase 1.2 and 3.9 multiple,and the protein expression was increased 0.9 and 4.7 multiple.Conclusion A high expression of survivin was induced in HepG 2 by 5 Fu,and resist apoptosis induced by chemotherapy agent.
