摘要
目的 探讨生长抑素类似物奥曲肽对人肝癌细胞产生抑制作用及其抑制机制。方法 以奥曲肽作用于人肝癌细胞株 SMMC- 772 1细胞 ,以人成纤维细胞株 HF为正常细胞对照 ,以 5 - Fu为阳性药物对照 ,通过噻唑蓝比色实验 (MTT实验 )、细胞增殖生长反应及传代实验和流式细胞术分析 ,得出结论。结果 在体外 ,奥曲肽对 SMMC- 772 1细胞产生抑制作用 ,该抑制作用具有量效关系和饱和性。奥曲肽与 5 - Fu对 SMMC- 772 1的抑制程度无明显差别 ,且对 HF细胞无影响。结论 奥曲肽对 SMMC- 772 1细胞具有抑制作用 ,诱导肿瘤细胞凋亡是其作用机制之一。
Objective The purpose of this study was to investigate the inhibition and the mechanism of octreotide,a somatostatin analogue on human hepatoma cell line in vitro.Methods Octreotide was added in human hepatoma cell line SMMC-7721,normal human fibroblast cell line HF and 5-Fu were used as control.MTT assay and the reaction of cell proliferation combined with flow cytometry were performed in this study.Results Octreotide can inhibit the growth of SMMC-7721 in vitro with certain concentration.There were no different between octreotide and 5-Fu in their inhibition on SMMC-7721.Octreotide has no effects on normal human fibroblast cell line HF.When SMMC-7721 were treated with octreotide,the typical apototic peak was identified by flow cytometry.Conclusion We conclude that octreotide can inhibit the proliferation of human hepatoma cell line SMMC-7721 in vitro,The induction of apoptosis by octreotide might be the primary mechanism.
