摘要
目的 建立大鼠主动脉平滑肌细胞体外培养模型 ,为动脉粥样硬化等心血管疾病的发病机制的研究提供了重要手段。方法 将大鼠主动脉翻转 ,两端结扎 ,用 0 .1%胶原酶消化血管内皮细胞 ,剩余血管剪成小块均匀种植于培养瓶底 ,加入DMEM培养液培养 ,观察血管平滑肌细胞的生长。结果 平滑肌细胞 4 8h后贴壁生长 ,早期呈多角形 ,以后呈梭形伸展。胰酶消化后平滑肌细胞可传 6~ 7代。结论 此方法简单易行 ,是获得血管平滑肌细胞的一种可靠方法。
Objective To establish a vascular smooth muscle cell(VSMC) model of rat in vitro. Methods Thoracic aorta of Wistar rat is obtained,by using a specially made crochet hook, the aorta is over turned,then ligated the two ends,the vascular endothelial cells are wiped off with 0.1% collagen I enzyme for 15 minutes. The aorta is divided into some little parts that is eventually paved on the bottom of cultivation flask with DMEM immerged its base.Cells are cultivated for 48 hours without movement in the condition of 5% CO 2 at 37℃. Results The initial VSMC can be found crawled from the edges of the vascular parts in form of shuttle. Conclusion This is an convenient way to obtain VSMC for study.
