Effects of Different Doses of Angiotensin Ⅱ on Proliferation and Intracellular Free Calcium Content of Rat Hepatic Stellate Cells

摘要 目的观察不同浓度血管紧张奏Ⅱ (Ang Ⅱ )对大鼠肝脏星状细胞 (HSC) [Ca2 +] i 及其增殖的影响 ,以探讨HSC和细胞内 [Ca2 +] i 的相关性。 方法HSC分离培养后 ,采有MTT法和Fura -2 AM荧光指示剂负载分别测定HSC增殖情况及细胞内 [Ca2 +] i。 结果 10 - 9~ 10 - 5mol LAngⅡ能显著增加HSC内游离 [Ca2 +] i(P <0 .0 1) ,且呈剂量依赖关系 ;10 - 9~ 10 - 6mol LAngⅡ均能促进HSC增殖 ( P <0 .0 5 ) ,但 10 - 5mol LAngⅡ却对HSC增殖无显著影响 (P >0 .0 5 )。 结论在一定剂量范围内Ang Ⅱ能够剂量依赖性地促进HSC增殖和增加HSC内游离 [Ca2 +] i,二者存在一定相关性。 Objective To investigate the effects of different doses of angiotensin Ⅱ(Ang Ⅱ) on proliferation and concentration of intracellular free calcium of rat hepatic stellate cells(HSC) and to study the correlation between proliferation of HSC and intracellular free calcium content. Methods HSC were isolated and cultured. Proliferation and intracellular free calcium concentration were measured by the MTT method and flurescence measurement with Fura-2/AM. Results It showed that 10 -9 ~10 -5mol/L Ang Ⅱcould significantly increase intracellular free calcium content of HSC in a dose-dependent style;10 -9 ~10 -6mol/L Ang Ⅱ could significantly promote proliferation of HSC(P<0.05),but 10 -5mol/L Ang Ⅱ could not significantly promote proliferation of HSC(P>0.05). Conclusion Ang Ⅱ could increase intracellular free calcium content in a dose-dependent style and promote HSC proliferation which being certain in the range of appropriate dose, correlativity.
