摘要
目的研究辛伐他汀对兔粥样硬化髂动脉肿瘤坏死因子 -α(TNF -α)表达的影响。 方法 30只雄性新西兰大白兔 ,分别予普通饲料喂养 (n =8)及高脂喂养 (胆固醇 1g d ,猪油 10 g d ;n =2 2 ) 6周。高脂喂养 2周后行髂动脉内膜球囊损伤术 ;术后辛伐他汀组 (n =12 )予辛伐他汀 15mg d ;分组喂养 4周后处死所有动物取一侧病变髂动脉做病理切片 ,用免疫组化方法观察TNF -α的表达情况 ;取另一侧病变髂动脉抽提总RNA和总蛋白 ,分别应用半定量逆转录多聚酶链式反应和Western蛋白印迹测定TNF -αmRNA和蛋白质水平的表达。 结果辛伐他汀可以显著降低兔血浆总胆固醇 ,降低粥样硬化髂动脉组织TNF -α的表达 ,正常对照组、基础对照组和辛伐他汀组动脉壁TNF -αmRNA的相对值分别为 0 .16± 0 .0 2 4、0 .5 6± 0 .0 5 0、0 .37± 0 .0 31,相互间有统计学差异 (P<0 .0 5 ) ,Westernblot检测和免疫组化结果一致。 结论辛伐他汀可以显著降低兔粥样硬化髂动脉组织TNF -α的表达。
Objective To investigate the effect of simvastatin on expression of tumor necrosis factor-α (TNF-α) in the atherosclerotic iliac artery of rabbits. Methods Thirty male New Zealand white rabbits were studied. The rabbits were fed on a normal diet(n=8) or a cholesterol diet (cholesterol:lg/d,lard: 10g/d, n=22) for 6 weeks. The iliac arteries of the cholesterol diet rabbits were balloon injury 2weeks after initiation of the diet, and twelve of them were fed with simvastatin (15mg/d) after in jury four weeks afterwards , untreated and balloon injury iliac arteries were harvested for immunocytochemical staining, Western blot, and semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) studies,respectively. Results The relative values of arterial TNF-α mRNA in 3 groups were 0.16±0.024,0.56±0.050,0.37±0.031, respectively with significant difference among them (P< 0.05). The expression of TNF-α antigen in the simvastatin group was reduced compared with the baseline group. Conclusion Simvastatin can reduce excessive expression of TNF-α in atherosclerotic iliac arteries.
