摘要
利用一株热带假丝酵母突变株SP -UV -5 6在 2 0m3 发酵罐中进行十三碳二元酸发酵的工业试验研究。采用等功率放大准则 ,确定了搅拌功率和搅拌速率 ;考察了搅拌器桨型对产酸的影响 ,采用六平叶涡轮搅拌器试验 ,连续发酵 3批次 ,培养 6d的平均产酸量为 15 6 .5 g/L ,较中试和小试的产酸量分别提高了 10 .2 %和 8.7%,最高产酸量达到 172 g/L ,与六箭叶涡轮搅拌器相比平均产酸量增长了 15 .5 %,但能耗增加 15 .6 %。选择一种有机溶剂 ,采用重结晶方法精制产物 ,产品中总二元酸质量分数达 99%以上 ,其中十三碳二元酸达 97%以上。
Fermentation of 1,13-tridencane dicarboxylic acid(DCA 13)was carried ont in an industrial pilot plant with a 20 m 3 stirred tank.Geometric similarity and equivalent power-per-unit-volume (P 0/V =2.0 kW/m 3) were applied for the scale-up. The main process parameters were calculated for designing a 20 m 3 stirred tank. A Candida tropicalis mutant SP-UV-56, which hardly assimilates n-alkanes as carbon source but assimilates sucrose, and optimized media and fermentation conditions were employed to manufacture DCA13 from n-tridecane(n-C13) as transformation substrate. Effects of two types of impellers such as flat blade disc turbine and arrow-like flat disc turbine on DCA13 yield were investigated. The average yield for the former was 156.5 g/L and 135.5 g/L for the latter after 144 h cultivation. A 15.5% increase in yield and a 15.6% higher power consumption were observed. The yield was 10.2% and 8.7% higher than that with 10 L and 3 m 3 fermenters respectively. The highest yield reached 172 g/L in a stirred tank installed with flat blade disc turbine impellers. A white powder product containing over 99% of total precipitated dicarboxylic acids with over 97% of DCA13 was obtained by recrystallization using an organic solvent. Specifications of DCA13 were reported.
