摘要
目的探讨细胞免疫在实验性自身免疫性肾小管间质性肾炎中的作用。方法兔TBM、完全福氏佐剂和百白破疫苗免疫后的脾细胞 (3 5~ 7× 10 8)静脉注射到正常Wistar大鼠 ,并行病理学及免疫组化检查。结果免疫后的脾细胞静脉注射后 ,于 14天发现Wistar大鼠发生TIN ,表现为单个核细胞浸润 ,大多数为T细胞 (占 6 8 3%) ,其中主要为辅助T细胞 (W3 /2 5阳性细胞 ,占 5 3 5 %) ,少数为抑制T细胞 (OX— 8阳性细胞 ,占 10 8%) ,还可见少量散在多核巨细胞。浸润的炎细胞几乎均表达MHCⅡ类抗原 (OX 6阳性 ) ,TBM上未见IgG沉积。结论细胞免疫在此类动物的TIN发生发展中起着重要的作用。
Objective To study the role of cellular immunity in experimental autoimmune tubulointerstitial nephritis. Methods After splenic cells from Wistar rats immunized with rabbit TBM and adjuvants were injected intravenously into normal Wistar recipients, characterization of renal mononuclear cells were detected by using immunohistochemistry. Results Intravenous transfer of splenic cells to naive Wistar rats resulted in TIN. Among the infiltrated mononuclear cell, most are T cells (68.3%), which of those cells mostly are TH cell (W3/25 positive cells, 53.5%). Characterization of the inflammatory infiltrates in the recipient kidney showed virtually all cells stained with OX-6. Immunofluorescence revealed no IgG fixation to TBM. Conclusion This model strongly suggests an important role for cell-mediated immunity in TIN in this strain.
