CpG序列及阳离子脂质体对猪带绦虫抗原基因免疫应答的影响 被引量:6

Regulating Effect of CpG Motifs and Cationic Liposome on Immune Responses to Antigenic Gene of Taenia Solium in Mice

摘要 本实验将猪带绦虫抗原基因VTS76分别与pUC1 8质粒、重组pUC1 8 CpG质粒联合及阳离子脂质体包装后免疫小鼠。结果表明 :pUC1 8和 pUC1 8 CpG能显著提高免疫小鼠总IgG和特异性抗体水平 ,增加小鼠免疫细胞数量 ,增强淋巴细胞增殖活性及白细胞介素 2的诱生活性。其中 ,pUC1 8 CpG的免疫增强效果更显著。阳离子脂质体包裹具有减少质粒用量。 Experiment was conducted to investigate the regulating effect of pUC18 plasmid, pUC18 CpG plasmid, and entrapment of pUC18 plasmid and pUC18 CpG plasmid on immune responses of mice co inoculated with antigenic gene of Taenia solium (VTS76). The results indicated that the IgG content and titre of specific antibodies to VTS76 mounted remarkably in the sera from the pUC18 and pUC18 CpG vaccinated mice; numbers of white blood cells, monocytes, granulocytes and lymphocytes significantly increased respectively in the mice vaccinated with pUC18 and pUC18 CpG plasmid, compared with those of the control; the proliferation of lymphocytes and induced IL 2 activities were significantly enhanced in the vaccinated groups. The above mentioned immune responses of mice co inoculated with pUC18 CpG were significantly stronger than those of co vaccinated with pUC18. The entrapment of these plasmids with cationic liposome could not only decrease the dosages of plasmids but also significantly increase levels of cellular and humoral response of mice.
