摘要
从分子水平上研究了肝片吸虫的抗原组分 ,用制备性 SDS- PAGE纯化 ES18.6 k D蛋白抗原 ,并用它成功制备了单因子兔抗血清 ,用这种抗血清来分析肝片吸虫各部分抗原 ,证实各部分抗原的 18.6 k D多肽具有相同的抗原决定簇 ,而且这种抗原决定簇不存在于其它分子量的多肽中。体抗原 (BA)和分泌排泄抗原 (ES)的双向电泳(Two- dimention Electrophoresis,2 - D)分析研究表明 ,肝片吸虫同一种分子量的蛋白质大多由多种等电点不同的多肽组成 ;同时也显示了体抗原和分泌 -排泄抗原分子量相同的蛋白质所包含的多肽数量和性质是有差异的。
Extracts of F. hepatica-excretory secretory products(ES), and body extracts (BA) of adult worm were analysed by protein electrophoresis (Two-dimention electrophoresis),and the antigen components of Fasciola hepatica was analysed upon a molecular level.The paper reported one monospecific antiserum(rabbit) prepared with 18.6kD peptide of ES purified by preparing SDS-PAGE, and its immunoblotting with four parts of extracts. The results showed 18.6kD in four parts of extracts had the same antigen determinant, while other peptides in different M.W. didn't.Two-dimention electrophoresis (2-D) of ES and BA indicated that many peptides in different PI had same M.W.,and differences were observed between ES and BA.
