摘要
目的研究神经生长因子(NGF)对Aβ25-35诱导海马细胞毒性的防治作用。方法取新生3d内大鼠海马细胞行体外培养7d,分别在加入Aβ25-35前后加入NGF和等体积培养液,采用MTT和β-tublin、ChAT免疫组化检测。结果β-tublin免疫组化证实所培养的为特异性结构完整的海马神经元。MTT及免疫组化显示Aβ25-35导致细胞活力显著降低及ChAT免疫阳性细胞明显减少。结论NGF能有效地阻止和轻度地修复Aβ25-35诱导的海马细胞损伤,对阿尔茨海默病的防治提供了一定的实验依据。
Objective To observe the protective and therapeutic effects of nerve growth factor (NGF) on β Amyloid25 35(Aβ25 35)-induced hippocampal neuron injury.Methods Hippocampal neurons obtained from 15 new born SD rats within 3 days were cultured in neural basal medium+B27 factor for 7 days,then Aβ25 35(10μg/ml) was added before and after addition of NGF(10μ/ml) and equal amount of culture fluid separately in two groups.MTT detection and β tublin &ChAT immunohistochemistry were used to determine the integrity of hippocampal neurons,cell viability and ChAT positive neurons.Results β tublin immunohistochemistry demonstrated the integrated hippocampal cells.MTT and ChAT immunohistochemistry showed that the viability of hippocampal cells and the number of ChAT positive neurons were decreased significantly in Aβ25 35 treated group.Conclusion NGF can effectively prevent and mildly improve the injury of hippocampal neurons induced by Aβ25 35.NGF may have protective and therapeutic effects for Alzheimer's disease.
