摘要 试验研究了超低温贮存顽拗型(RecaIcitrant)种子茶籽的技术,结果表明,采用铝箔/薄膜复合袋密封包装,将含水量8%至13.83%的种子直接快速投入液氮,35℃温水解冻方式,能保持生活力,最适含水量为13.83%,经118天贮存后发芽率93.3%,且均成苗,液氮贮存后的最适发芽方法是在5%水分(W/W)沙床中于5±1℃预吸处理15天,然后移至25℃发芽,预处理后的种子,细胞膜修复能力增强,渗漏物减少,发芽率提高。 The technique of cryopreservation(-196℃)of recalcitrant tea(Camellia sinensis L.)seeds was investigated.Tea seeds of moisture content from 8% to 13.83%(wet weight)were sealed in laminated aluminum foil packets and stored in liquid nitrogen(LN2)for different periods.The tea seeds were immersed directly and rapidly in LN2 from room temperature and rewarmed rapidly in 35℃ warm water.Under such condition,the viability remained well,and the most suitable moisture content was 13.83%,both germination percenlage and emergence were 93.3% after storage in LN2 for 118 days.The suitable method of germination lest for tea seeds stored in LN2 was 25℃ after preconditioning in 5% MC(w/w)sand bed at 5±1℃ for 15 days.The ability of membrane repair increased,solute leakage reduced and germinability improved after pretreatment of cryopreserved seeds.
