摘要
目的 :探讨性激素单独应用或联合应用对人脐静脉内皮细胞释放一氧化氮 (NitricOxide ,NO)的影响。方法 :取健康产妇分娩后 6h内的脐带内皮细胞进行培养 ,以Griess测定培养液中NO2 含量间接反映NO生成量 ,分别测定加入 0 .0 0 1μmol/L ,0 .1μmol/L和 10 μmol/L的 17β E2 ,P、T刺激细胞 10min、30min、6 0min时NO2 含量 ,以 1ml无钙镁盐液作对照。结果 :与对照组相比 ,17β 雌二醇浓度为 0 .0 0 1μmol/L、0 .1μmol/L、10 μmol /L单独作用于内皮细胞 10min和 30min时 ,培养液中NO含量明显增加 (P <0 .0 5 ) ,但刺激 6 0min时无明显作用 (P >0 .0 5 ) ;与孕激素 (0 .0 0 1μmol/L、0 .10 μmol/L、10 μmol /L)分别联合应用 ,作用于内皮细胞 10min、30min和 6 0min ,都能显著抑制雌激素刺激内皮细胞释放NO(P <0 .0 5 )。不同浓度和作用时间下 ,孕激素和雄激素对内皮细胞释放NO的作用不同。结论 :雌激素能够通过NO途径产生血管舒张作用 ,孕激素可能通过抑制NO的释放而抑制雌激素舒张血管的作用。
Aim: To explore the influence of individual or combined use of sex hormones on NO release from endothelial cells in human vena umbilicalis. Methods: Primary endothelial cells in human vena umbilicalis were cultured, and the NO 2 contents in culture solution were determined by enzyme process. Results: In comparison with controls, NO contents significantly increased when 17β-estradiol ( 0.001 μmol / L, 0.1 μmol / L, and 10 μmol / L) were administered to the endothelial cells for 10 min and 30 min, respectively (P<0.05), but treatment with 17β-estradiol for 60 min caused no significant changes of NO (P>0.05). Combined with progesterone ( 0.001 μmol / L, 0.1 μmol / L, and 10 μmol / L), the effects of 17β-estradiol on the NO release from endothelial cells were markedly inhibited(P<0.05). The actions of progesterone and testosterone on the NO release from endothelial cells were different with different concentrations and time of administration. Concluson: 17β-estradiol dilates the blood vessel,which may act via mechanism of affecting the release of nitric oxide from endothelium cells.When 17β-estradiol and progesterone are combinedly used,progesterone inhibits the effect of estradiol on the vasodilatation possibly by inhibiting the release of NO
