同型半胱氨酸和金属硫蛋白对血管内皮细胞的影响

摘要 目的 :观察同型半胱氨酸 (Hcy)血症时小鼠肝、心、肾组织金属硫蛋白 (MT)含量的变化 ,以及MT对Hcy损伤内皮细胞的影响。 方法 :①昆明种小鼠 12只 ,分为对照组和高Hcy组 ,高Hcy组小鼠腹腔注射Hcy(16μmol/kg) ,复制高Hcy血症模型 ,对照组同法注入等量生理盐水。以Cd 血红素饱和法测肝、心和肾组织MT水平。②培养人血管内皮细胞 ,分为空白对照组 ,单用Hcy(1mmol/L)组及MT(2× 10 -5mol/L)和Hcy(1mmol/L)同时应用组 ,孵育人血管内皮细胞 6h测定细胞存活率、培养液乳酸脱氢酶 (LDH)活性和蛋白漏出量。结果 :Hcy组小鼠肝、心和肾组织MT水平较对照组分别增加 2 10 %(P <0 .0 1)、133%(P <0 .0 5 )和 6 0 %(P <0 .0 1)。MT +Hcy组培养的内皮细胞存活率较Hcy组培养的内皮细胞升高 13.9%(P <0 .0 1) ,培养液LDH和蛋白漏出量较Hcy组分别降低 2 7.1%(P <0 .0 5 )和 31.4%(P <0 .0 1)。结论 :Hcy诱导小鼠肝、心、肾组织MT生成 ,MT拮抗Hcy的细胞损伤作用。 Aim: To observe the changes of metallothionein (MT) in the liver, heart and kidney of mice during hyperhomocysteinemia and influences on endothelial cell damaged by Hcy. Methods: ①12 mice were divided into control group and Hcy group.Hyperhomocysteinemia model of mice was prepared by intraperitoneal injection of homocysteine (16 μmol/kg). The contents of MT in liver, heart and kidney tissues were determined. ②Endothelial cell was cultured by Hcy(1 mmol/L) and MT(2×10 -5 mol/L)+Hcy(1 mmol/L) respectively. We measured the number of viable cell in all group,tested the capacity of LDH and protein in cultured medium. Results: Compared with control group,MT levels in Hcy group were increased by 210%(P<0.01) for liver, by 133%(P<0.05) for heart and by 60%(P<0.01) for kidney tissue. Compared with that in Hcy group,the viability of cultured endothelial cell in MT+Hcy group is 13.9%(P<0.01) higher,the contents of LDH and protein in cultured medium respectively decreased 27.1%(P<0.05) and 31.4%(P<0.01). Conclusion: Hcy could induce the formation of MT in the liver, heart and kidney of mice, and MT could inhibite the damage of Hcy cells.
