Detection of Human Telomerase Reverse Transcriptase Messenger RNA in Urinary Cancer Tissue Samples

摘要 目的 探讨泌尿系肿瘤中端粒酶催化基因 (hTRT)mRNA的表达与肿瘤分期和分级的关系。方法 应用原位杂交技术对膀胱癌、肾癌标本和膀胱、肾脏正常组织进行hTRTmRNA表达检测。结果  48例膀胱癌标本中 42例hTRTmRNA阳性表达 ,hTRTmRNA阳性率为 88% ,对照组 30例中 2例正常膀胱组织黏膜阳性表达 ,两者存在明显差异 (P <0 .0 1) ,肿瘤分级与肿瘤组织hTRTmRNA表达情况无相关性。 2 9例肾癌组织hTRTmRNA阳性 2 5例 ,阳性率为 86 % ,对照组 30例中 2例肾脏正常组织hTRTmRNA阳性表达 ,两者存在明显差异 (P <0 .0 1) ,肿瘤分期与肿瘤组织hTRTmRNA表达情况无相关性。结论 hTRTmRNA在肾癌和膀胱癌中有异常阳性表达 。 Purpose: To evaluate the expression of human telomerase reverse transcriptase messenger RNA(hTRT mRNA) in urinary cancer tissue samples and the relationship with cancer stages and grades. Methods: We investigated the expression of hTRT in 48 cases of bladder cancer, 29 renal cancers, 30 bladder tissues and 30 renal tissues by in situ hybridization. Results: 42 bladder cancer cases expressed hTRT mRNA, while only 2 normal tissues had moderate expression of hTRT mRNA. The expression level of hTRT mRNA of bladder cancer was significantly different from normal bladder (P 0.05). 25 renal cancer cases expressed hTRT mRNA, while only 2 renal tissues had moderate expression of hTRT mRNA. The expression level of hTRT mRNA of renal cancer was also significantly different from normal renal tissues (P 0.05). Conclusions: Rneal and bladder cancer tissues have increased positive expression of hTRT mRNA, yet the expression has no relationship with cancer stages and grades.
