用新鲜白血病细胞或细胞株免疫BALB/c小鼠,经细胞融合技术制备得6株能稳定分泌抗人髓系细胞分化抗原单克隆抗体(以下简称单抗)的杂交瘤株.经鉴定该组单抗各自所识别的特异性抗原分子量为46~150kd.这些抗原在粒单系细胞膜上均有程度不等的表达,6种单抗中有5种具有结合兔补体的能力,其对白血病细胞株补体依赖的特异性细胞毒为73%~94%.两种单抗(Zh_(820),Zh_(114))对正常CFU-GM生长有抑制作用外,余4种单抗无明显影响.6种单抗均与FAB M_1~M_5型急性髓系白血病细胞有不同程度的阳性反应.
Using the hybridoma technology, the author produced and applied six monoclonal antibodies (McAb) against human myeloid cells. These McAbs were produced by immunizing BALB/c mice with whole cell suspensions of fresh AML cells or HL-60 cells and were named Zh_(53), Zh_(2-1), Zh_(805), Zh_(826) Zh_(820) and Zh_(114), respectively. Western blotting analysis showed that these McAbs could recognize different antigens with different molecular weights. These antigens expressed themselves on the surface of leukemic cells from most patients with acute myeloid leukemias and some normal human myeloid cells. Five of the six McAbs were capable of fixing rabbit complement and showed strong complement-mediated cytotoxicity to leukemic cell line HL-60 cells with dead cells ranging from 73% to 94%. CFU-GM growth assay showed that the complement-fixing antibodies Zh_(53), Zh_(2-1) and Zh_(805) did not produce any significant inhibition to normal CFU-GM colony growth, but the antibodies Zh_(820) and Zh_(114) revealed their respective inhibition rates of 24% and 20%.
