聚焦超声无创治疗阿尔茨海默病研究进展

摘要 近年来阿尔茨海默病研究取得了巨大进展,但仍缺乏有效治疗方式。目前几种针对病理标志物的靶向疗法的热点研究,虽能改善症状却无法阻止疾病进程。由于聚焦超声技术可以无创、可逆的打开局部血脑屏障,联合微泡可促进脑部对药物的吸收,单独超声辐照可显著降低实验动物脑部淀粉样蛋白斑块水平并改善病症症状,特别是聚焦超声打开血脑屏障Ⅰ期临床实验成功,展示了此项无创治疗技术用于阿尔茨海默病治疗具有巨大潜力。本文对近十年聚焦超声技术治疗阿尔茨海默病的研究进展进行综述。 Despite decades of advances in Alzheimer’s disease(AD),there remain no effective treatments that significantly alter its natural history.Currently,there are several treatments targeted the proteins occurs in its pathophysiology,but most of them are proved to be not effective and can not alter the course of the disease.Recent work investigating using focused ultrasound in transgenic animal models has shown that focused ultrasound coupled with microbubble or ultrasound alone can facilitate drug delivery thus reducing plaque burden and reversing memory deficits.With the success of phaseⅠclinical trail of opening brain blood barrier using focused ultrasound,this image-guided,noninvasive technique appears promising.This article reviews the current research progress of therapy of Alzheimer’s disease using focused ultrasound.
