摘要
从 5个不同甘蓝型油菜品种 (系 )的下胚轴分离得到游离原生质体 ,以改良KM 8p原生质体培养基作液体浅层暗培养。结果表明 ,低温 (15℃ )暗处理培养无菌苗的下胚轴原生质体活力达 92 .1% ,培养 7d后的细胞分裂频率达 2 6 .2 %。分化培养基附加 5mg/L 6 -BA ,再生频率可达 17.6 %。利用这一培养体系 ,5个油菜品种均获得再生植株。
The vigour of the protoplasts isolated from hypocotyl tissue under the seeds germinated at 15℃ in the darkness of 5~7 day old seedlings of Brassica napus was 92.1%. The protoplasts cultured in a modified KM8p medium with 'thin liquid layer' method, had a frequency of cell division of 26.2% after being cultured for 7 days. The frequency of shoot regeneration was 17.6% on differentiation medium supplemented with 5mg/L 6-BA. Using this procedure, regenerated plants were obtained in five rapeseed varieties.
