摘要
用CFDA/AM荧光探针负载离体培养的大鼠心肌细胞 ,在激光扫描共聚焦显微镜下观察细胞内胞间通讯的变化。结果发现不同浓度和时间的FGF - 2对原代培养的新生大鼠的心室肌细胞的胞间通讯的影响不同 ,lng/m1组在FGF— 2作用第l天和第 4天对细胞的胞间通讯无明显变化影响 ;10ng/ml和 10 0ng/ml在FGF— 2作用的第 1天的平均荧光恢复速率明显下降 ,在作用后的第 4天 ,平均荧光恢复速率依然显著低于对照组。 10 0 0ng/ml的FGF - 2作用于心肌细胞的第 1天 ,平均荧光恢复速率无显著变化 ,第 4天平均荧光恢复速率则明显增加。
Cultured cardiocyte were loaded with CFDA/AM fluorescence probes, and the changes of cell—cell communication were examined by laser scanning confocal microscopy. The results showed that the 1day after FGF-2 exertion on the cells, the mean fluorescence recovery rate were decreased in the groups of FGF-2 10ng/ml and 100ng/ml,while the changes in groups 1ng/ml and 1000ng/ml showed no statistic significance. 4days after FGF-2 exertion on the cells, the mean fluorescence recovery rate remained decreased in groups 10ng/ml and l00ng/ml, the mean fluorescence recovery rat in groups 1ng/ml again showed no significant changes, but in the groups 1000ng/m1 the mean fluorescence recovery rat increased.
