压电免疫传感器检测日本血吸虫循环抗原的研究 被引量:3

摘要 通过葡萄球菌蛋白A(SPA)将从感染兔血清 (InRS)或免疫兔血清 (ImRS)中纯化的IgG抗体固定于镀金的石英晶片上 ,制成一种新型的液相压电免疫传感器 ,用于检测感染兔血清中的日本血吸虫循环抗原 (SjCAg)。分别比较了提纯前血清、部分纯化抗体和完全纯化抗体 ,以及感染和免疫兔血清纯化IgG的固定效率和检测效果。结果表明 ,以完全纯化抗体的效果最好 ,免疫兔血清纯化IgG优于感染兔血清纯化IgG。同时 ,还研究了SPA的固定浓度、检测时间、血清最适稀释度及晶体的再生性等。用该法检测了不同感染度的兔血清 。 A new liquid piezoelectric immunosensor(PEIS) was developed to detect Schistosoma japonicum circulating antigens(SjCAg) in infected rabbit sera by immobilizing the purified IgG antibodies from infected rabbit sera(InRS) or immunized rabbit sera(ImRS) onto the golden electrode of piezoelectric quartz crystal through SPA The effect of the different kind antiserum,including InRS and ImRS and the purity of IgG,including crude sera,partially purified and fully purified IgG,were compared in the efficiency of immobilization and the detection of SjCAg by PEIS It was demonstrated that the immunized rabbit sera was better than the infected rabbit sera,and the fully purified IgG was the best among samples with different IgG purity.The concentration of SPA for immobilizing antibodies,the reaction time,the optimum dilution of detected sera and the regeneration of crystal were also observed Rabbits'sera with various degree of infection were examined It was found that frequency change was directly proportional to the degree of infection in a certain range
