摘要
目的 用淡色库蚊溴氰菊酯抗性相关胰蛋白酶核酸探针检测淡色库蚊溴氰菊酯抗性。方法 用逆Northern鉴定抗药性品系和敏感品系中胰蛋白酶的表达差异 ,用随机引物法标记鉴定后的胰蛋白酶cDNA片段为探针Northernblot检测淡色库蚊的抗药性。结果 逆Northern显示胰蛋白酶在抗药性品系中的表达量是敏感品系的 4 33倍。以其为探针Northernblot检测显示 ,胰蛋白酶基因在抗药性品系中高表达 ,抗药性品系中胰蛋白酶的表达量是敏感品系的 3 90倍。结论 作为一种新的抗药性检测的分子生物学方法 。
The difference expression of trypsin gene between deltamethrin resisitant strain and susceptible strain of Cx pipiens pallens was further investigated,and the results showed that the expression of trypsin gene was 4 3 fold more in the resistant strain than in the susceptible strain,by reverse Northern blot Then we used 32p labeled trypsin gene which had been labeled by random primer method as northern blot probe to detected the deltamethrin resisitance in Cx pipiens pallens It showed a 3 9 fold more expression in resistant strainthan in susceptible strain
