产黄青霉菌原生质体形成、再生和融合

摘要 采用0.5%的纤维素酶和0.5%的蜗牛酶的混合液在33°c分别酶解产黄青霉菌的两株营养缺陷型突变体的菌丝体以制备原生质体,并以0.6MNaCl为渗透压稳定剂使原生质体在再生培养基上再生成菌落,以30%的聚乙二醇(分子量6,000)和0.01MCaCl_2作助融剂,获得了两株营养缺陷型突变株原生质体营养互补的融合子,融合率为0.011% Protoplasts were prepared by the action of mixed enzymes system cont- aining cellulase and snail digestive juice preparation at 33℃ on the mycelium of two Penicillium auxotrophs. Purified protoplasts were incubated on solid medium containing 0.6M NaCl as smotic stability, and colonies were formed. Fusion of protoplasts was formed by 30% polyethylene glycol MW 6,000 0.01 M CaCl_2 as fusogenic agents. The fusion frequency was 0.011% of the number of protoplasts surviving the fusion treatment
