阿魏酸对脂多糖诱导的小鼠急性肺损伤的保护作用 被引量:3

摘要 探讨阿魏酸对脂多糖(LPS)诱导的小鼠急性肺损伤(ALI)的保护作用。将60只BALB/c小鼠随机分为6组,每组10只:正常对照组,模型组,阿魏酸高(30 mg/kg)、中(15 mg/kg)、低剂量组(7.5 mg/kg),地塞米松组(2 mg/kg)。灌胃给药,每天1次,连续7 d,末次给药2 h后除正常对照组小鼠气管注射等量生理盐水外,其他各组小鼠气管滴注脂多糖(10 mg/kg),6 h后眼球采血,处死小鼠,取新鲜肺组织,ELISA检测血清TNF-α、IL-6、IL-1β含量,计算肺组织湿/干重比值,观察肺组织的病理变化。结果显示,与正常对照组比较,模型组小鼠肺组织湿/干重比值、血清TNF-α、IL-6、IL-1β含量均显著升高,差异具有统计学意义(P<0.05)。与模型组比较,地塞米松组和阿魏酸高、中、低剂量组小鼠肺组织湿/干重比值、血清TNF-α、IL-6、IL-1β含量,均有不同程度降低,差异具有统计学意义(P<0.05)。模型组肺组织可见肺泡间隔增厚,大量炎性细胞浸润,部分肺泡腔缩小,含有较多的炎性细胞,地塞米松组和不同剂量阿魏酸组上述变化明显减轻,仅见肺泡间隔少量炎性细胞。结果表明,阿魏酸对脂多糖诱导的小鼠急性肺损伤有一定的保护作用,其机制可能与抑制体内炎性介质释放有关。 To study the protective effect of ferulic acid on acute lung injury(ALI)induced by lipopolysaccharide in mice.Sixty BALB/c mice were randomly divided into 6 groups,10 mice in each group:control group,lipopolysaccharide model group,high-dose ferulic acid group(30 mg/kg),medium-dose group(15 mg/kg),low-dose group(7.5 mg/kg),and dexamethasone group(2 mg/kg).Each group received continuous irrigation for 7 days,1 day 1 time,2 h after the everyday last delivery,All groups of mice,except the model group,received trachea injection of lipopolysaccharide(10 mg/kg)to establish a model group;the control group was given the trachea injection of same volume normal saline.6 h after modelin,mice were sacrificed to take fresh lung tissue,eyeball blood.Enzyme-linked immunoassay(ELISA)was used to detect serum TNF-α,IL-6,IL-1βlevels.The wet/dry weight ratio of lung tissues in mice was calculated.The pathological changes of lung tissues in each group were observed.Compared with the normal control group,the wet/dry weight ratio in the lung tissues of the mice were significantly increased and the serum TNF-α,IL-6 and IL-1βlevels in the lipopolysaccharide model group,dexamethasone group and the high-dose,medium-dose and low-dose ferulic acid groups were significantly increased(P<0.05).Compared with the lipopolysaccharide model group,the serum TNF-α,IL-6 and IL-1βlevels in the dexamethasone group and the high-dose,medium-dose and low-dose ferulic acid groups,as well as the wet/dry weight ratio in the lung tissues of the mice were significantly reduced to different degrees(P<0.05).The lung tissue of the model group showed thickening of the alveolar space,a large number of inflammatory cells infiltrated,some of the alveolar spaces were reduced,and there were more inflammatory cells.The above changes in the dexamethasone group were alleviated,and in the ferulic acid,the low,the middle and high dose groups,the above changes were alleviated and only a small amount of inflammatory cells in the alveolar space were found.Ferulic acid has a protective effect on Lps-induced acute lung injury to some extent,and its mechanism may be related to inhibiting the release of inflammatory mediators in vivo.
