摘要
目的观察胡椒碱对急性缺血性脑损伤大鼠的神经保护作用。方法随机将SD大鼠分为假手术组、生理盐水组、胡椒碱组、溶媒组,每组8只。采用颈内动脉线栓法制备大鼠大脑中动脉闭塞脑缺血模型,术后0.5 h灌胃给药,1次/d,连续3 d。不同时间点进行神经功能评分。72 h断头取脑,TTC染色,测量梗死体积。另择32只大鼠,分组同上,于脑缺血24 h处死,断头取脑,观察神经细胞凋亡及Caspase-3蛋白表达。结果胡椒碱组大鼠神经功能评分、脑梗死体积、凋亡细胞数及Caspase-3阳性细胞数较生理盐水组、胡椒碱溶媒组降低,差异有统计学意义(P<0.05)。结论胡椒碱通过减少Caspase-3蛋白的表达抑制细胞凋亡、缩小脑梗死体积、改善神经功能。
Objective To investigate the neuroprotective effect of piperine on acute ischemic brain injury in rats.Methods Thirty-two SD rats were randomly divided into four groups,namely shamed group with saline,saline group with saline,vehicle group with vehicle and piperine group with piperine.The MCAO model was established with suture occlusion technique according to modified Zea Longa method.The neurologic impairments were evaluated at 2 h,24 h,48 h and 72 h after operation of MCAO.All of the rats were killed at 72 h.The infarct vol-umes were calculated by TTC staining.The other 32 male SD rats were treated the same as above.The rats were killed at 24 h after MCAO.The apoptosis cell and expression of caspase-3 were detected.Results The neurological deficit scores of piperine group were signifi -cantly decreased compared with saline and vehicle group.The piperine could reduce the infarcted volumesof the apoptosis cells and caspase-3 positive cells(P <0.05).Conclusion The piperine can decrease the infarcted volumes and inhibit the apoptosis of cells by reducing the ex-pression of caspase-3.
