裂殖壶菌丙二酸单酰转移酶(MAT)基因的克隆与序列分析

摘要 二十二碳六烯酸(DHA)是一种重要的多不饱和脂肪酸,裂殖壶菌(Schizochytrium)因富含DHA成为当前的研究热点。丙二酸单酰转移酶(MAT)是该菌合成DHA的关键酶。以Schizochytriumsp.FJU-512cDNA文库中的EST序列contig417(GenBank accession No.EF483907)为基础,获得了丙二酸单酰转移酶基因的DNA序列和cDNA序列。序列分析表明,cDNA序列与DNA序列完全一致,开放阅读框全长1 176bp,编码391个氨基酸,且具备该类酶氨基酸序列的两个经典保守区。对该蛋白进行系统发育分析,构建进化树,结果显示,该蛋白同Schizochytriumsp.TIO01的丙二酰辅酶A-ACP转移酶(MCAT)亲缘关系最近。 Docosahexaenoic acid(DHA)is an important polyunsaturated fatty acid.Schizochytriumhas captured a research hotspot because of its abundant content of DHA.Malonyl CoA-acyl carrier protein transacylase(MCAT)is an essential component involved in the biosynthesis of DHA.In this study,the DNA and cDNA of a putative MCAT gene was observed based on the EST sequence contig417(GenBank accession No.EF483907).The putative MAT gene contained an 1 176 bp open reading frame encoding aprotein of 391 amino acids,exhibited two high novelty amino acid sequences.The result of phylogenetic analysis of the protein and the construction of pedigree indicated that the consanguinity of the protein was very close to MCAT of Schizochytriumsp.TIO01.
