成年狗股骨微波灭活骨缺损模型的建立及其修复 被引量:3

Development and Repair of Canine Femoral Defect Model Caused by Microwave-Induced Hyperthermia with Composite Material of Bone Morphogenetic Protein

摘要 目的 :建立成年狗股骨经微波灭活造成骨缺损的实验动物模型 ,并用复合骨形成蛋白材料修复。方法 :用自制的骨肿瘤微波治疗仪 ,以 1.5kHz、70W功率加热至 5 0℃~ 5 5℃ ,维持 2 0min ,造成狗股骨中段骨缺损 ,术后不同时段观察愈合情况。制备复合骨形成蛋白材料 ,经评价后植入骨缺损处修复并观察结果。结果 :在保持狗股骨连续性的前提下 ,长 2 .5cm、宽 1.0cm的骨缺损 9个月时无愈合倾向。复合骨形成蛋白材料具有成骨诱导活性和生物相容性、有粘合性和可塑型性 ,具备有利于新生血管长入的空间 ,可与宿主骨“生物铆定”。结论 :成年狗股骨微波造成的骨缺损实验模型 ,长 2 .5cm、宽 1.0cm 9个月不能自愈 ,适宜于各种骨修复材料的填充。复合骨形成蛋白材料修复微波灭活骨缺损效果良好 ,可作修复骨缺损的骨替代材料。 Objective:To develop a model of canine femoral defect caused by microwave induced hyperthermia and repair with the composite material of bone morphogenetic protein.Methods:The microwave machine self made for the treatment of bone tumor was used to cause the canine femoral defect of different size with frequency of 1.5kHz and power of 70W.The canine femur was heated to 50~55℃ and maintained for twenty minutes.The composite material of bone morphogenetic protein was prepared beforehand.After the examinations,the composite material was implanted into the canine femoral defect.The canine femurs were examined at different postoperative time.Results:With the continuity of the canine femur,the bone defect could not be healed self when it was 2.5cm of length and 1.0cm of width.The combined material could be remodeled easily and the new blood vessels could growth into because there were a large amount irregular gaps in it.New bone could be formed by the induction of bone morphogenetic protein and the decalcified bone matrix could be replaced by the new bone.So the composite material could be merged with the normal bone.Conclusions:The canine femoral defect caused by microwave induced hyperthermia with length of 2.5cm and width of 1.0cm can not be healed self and it is favorable for different materials to be implanted in.So it is an ideal experimental model and the composite material of bone morphogenetic protein is an ideal material for the repair of bone defect.
