组织工程学的回顾和未来的研究方向 被引量:1

摘要 组织工程学是一门新兴的学科,它应用细胞生物学和工程学的原理在体外或在体内制造组织和器官从而达到修复损伤组织和改善其功能的目的。组织工程学的提出和建立虽然只有10多年的时间,但其迅猛发展的势头展示出在未来医学发展中的重要作用。 组织工程的建立和发展 大约在十多年以前,美国哈佛大学外科医生Vacanti JP因器官供应来源的问题在大海边上瞑思苦想之际,碰到了麻省理工学院的化学生物工程师Robert Langer。Langer向Vacanti建议把细胞放在可吸收的合成材料中来形成组织或器官。这次偶然的海边相遇引发了组织工程在美国的兴起和在全世界的扩展和发展。1987年,美国国家科学基金会正式提出“组织工程”一词并确定这一门新兴学科的建立。 早期的组织工程主要集中在单一组织的构建。1988年,Vacanti等用分离的软骨细胞和可降解的生物材料首先在动物皮下构建成透明软骨,这一研究成果具有重要的意义。由于在体内,软骨损伤之后很难使透明软骨得到再生和修复,采用组织工程手段构建成具有功能的透明软骨无疑为将来的组织和器官的修复开辟了一条重要的途径。以后,Kim和Vacanti等又用PGA和PLA在裸鼠皮下构建成软骨组织。
