摘要
目的 研究兔颈动脉平滑肌细胞核DNA含量随缺血时间而变化的关系,判断断指再植时限。方法7只新西兰大白兔过量麻醉法处死,常温(25℃)和冷藏(0~4℃)保存条件下,在0h、4h、8h、12h、16h和24h时取材,Feulgen染色后,图像细胞光度仪检测颈动脉平滑肌细胞核DNA含量,统计学处理数据。结果平滑肌细胞核DNA含量在常温下缺血8小时后,随缺血时间延长而较有规律的下降,缺血12h以内下降不明显,12h后下降比较明显。冷藏组(0~4℃)平滑肌细胞核DNA含量无明显变化。结论 断指再植时限常温下以12h内为宜。冷藏保存的肢(指)体24h内都可再植。
Objective To evaluate the DNA degradation in smooth muscle cell nuclei of carotid in rabbits during ischemia, and study the relationship between the degradation and ischemic time. Then to definite the replantation time-limitation of severed digits at room temperature. Methods Seven New Zealand's rabbits were killed, and their corpses were deposited at room temperature and in refrigerator (0-4℃). Carotid section was acquired at Oh,4h,8h,12h,l6h,24h. DNA contents in nuclei of smooth muscle cells were evaluated by Feulgen reaction and image cytometry (ICM). Results The DNA content in nuclei of smooth muscle cells decreased at 8h after ischemia took place, and the decrease became significantly after 12h after ischemia happened. In refrigerated group, the DNA contents have no changed during 24 hours. Conclusion The time limitation of replantation of severed digits is 12h at room temperature. Preserved in refrigerator (0-4℃) ,serverd digits could be replanted when ischemic time is within 24 hours.
