摘要
研究利用自身酶和外源酶降解的技术方法对钝齿棒状杆菌菌体进行了处理。研究表明 :在 5 0℃ ,pH 6,菌体浓度 15 % ,反应 14h条件下 ,菌体自身降解的氨基酸态氮为 0 5 74% ,总核苷酸含量为 3 60mg/10 0mL。外源酶选择了 5 3 7酸性蛋白酶和由麦芽根提取的 5′—磷酸二酯酶 ,降解液的氨基酸态氮达到1 2 3 9% ,5′—单核苷酸为 82 2 8mg/ 10 0mL。
The technical methods of autolysis and hydrolysis of exogenous enzymes were used to process the protein and nucleic acid of Corynebacterium crenatium.The amino acid nitrogen(0 574%)and the total nucleotides(360mg/100ml)were produced by autolysis with 15%cell concentration at 50℃,pH 6 for 14 hours.When the acid proteinase 537 and 5′-phosphodiesterase extracted from the roots of malt were selected as the exogenous enzymes,amino acid nitrogen and 5′-nucleotide in the hydrolytic solution could reach 1 239% and 82 28mg/100ml,respectively.
