番木瓜悬浮培养的体胚发生与植株再生 被引量:14

摘要 番木瓜(Carica papaya L.)是一种在热带、亚热带地区广泛栽培的多年生果树。在医药和食品工业上有广泛用途。番木瓜组织培养工作中已有不少体胚发生的报道。Litz and Conover(1982,1983)在培养番木瓜胚珠时得到大量体胚发生并成苗。Chen等(1987)从来自根的愈伤组织得到体胚并再生植株。本文以番木瓜F_1代杂种小苗的根、茎为材料,通过悬浮培养途径,获得大量体胚并再生植株。 Compact embryogenetic calli were obtained from explants on P_3 medium after 4 weeks of culture and high-frequency somatic embryogenesis occurred after these calli were transferred into suspension culture. Experimental data showed that low level (0.2%W/V) of activated charcoal had beneficial effects on somatic embryogenesis. Abundant calli on P_4 medium however,showed no embryogenesis. On the other hand, callus induction and somatic embryogenesis varied with different varities of explants. The efficiency of somatic embryogenesis was much higher, if roots were used as explants, whereas stems were more suitable for callus formation Mature somatic embryos with cotyledons were cultured on MS medium containing different plant hormones. The optimum medium for germination and growth of entire plantlet was MS_0 medium. The somatic embryos on MS_2, MS and MS_3 media germinated rapidly, but formed excessive callus from the surface of germinating embryos.
