胞内共生菌Wolbachia内噬菌体相关尾蛋白基因的分子克隆及特征分析

摘要 Wolbachia是一种广泛存在于各种节肢动物体内经母系细胞质遗传的胞内共生菌.它能够改变宿主的生殖和发育行为,引起细胞质不亲和(cytoplasmic incompatibility,CI)、孤雌生殖、雌性化和雄性致死等现象,但是其间的分子生物学机理迄今尚不清楚.采用RDA(representational difference analysis,代表性差异分析)和LM-PCR(ligation-mediated PCR)等方法,通过研究Wolbachia不同品系基因组之间的差异,从来自果蝇Drosophila melanogaster CantonS的Wolbachia(wMelCS)中克隆到了噬菌体相关尾蛋白基因prtp(phage-related tail protein),同时从果蝇Drosophila melanogaster yw67c23中的Wolbachia(wMel)中克隆到了同源基因.通过研究prtp在不同Wolbachia品系间的表达,发现PrTP可能不直接参与CI过程.体外果蝇S2细胞的瞬时转染实验结果证明了prtp基因内双向核定位序列(bipartite nuclearlocalization signal sequence,NLS)的功能,prtp基因的存在为噬菌体介导的基因水平转移(horizontal genetransfer,HGT)提供了直接证据,并提示其在Wolbachia的生殖特性中可能扮演重要的角色.
