摘要
目的 观察造血干细胞移植 (HSCT)供受者HLA C分子结构差异 ,探讨免疫抑制信号系统在HSCT后GVHD中的作用。方法 利用DNA测序和蛋白质三维空间结构模拟分析方法回顾性观察HSCT后发生GVHD的供受者的HLA C分子结构差异。结果 HLA C分子在氨基酸序列、三维空间结构、电性分布和分子表面结构上有明显差异。结论 HSCT供受者间HLA C分子结构差异导致HLA C分子不能被供者免疫细胞的杀伤细胞抑制性受体识别 。
Objective To compare the difference of HLA C structures between donor and recipient after hematopoietic stem cell transplantation(HSCT),we investigated the impact of immunization inhibitory signals on graft versus host disease(GVHD)after HSCT.Method The difference of HLA C structures in a pair of donors and recipient in HSCT therapy with GVHD by detecting DNA sequecing and analyzing the domain structure of protein.Result There are obviously difference of HLA C structures between donors and recipients in amino acid sequences,three dimensional structures,electronic power and surface configuration.Conclusion The difference of HLA C structures between donors and recipients in HSCT resulted that HLA C of recipients cannot be identified by killer cell inhibitory receptor of donors,this may cause GVHD.
