白血病患者骨髓细胞双微体表达的细胞遗传学研究

Expression of double minutes in cases with leukemia

目的 探讨白血病患者骨髓细胞中双微体 (DM)的表达及与该病预后的相互关系。方法 对 32 0例白血病患者采用短期细胞培养法制作骨髓细胞染色体标本并观察DM表达。结果 ①在 32 0例患者中有 2 8例见DM表达 ,占 8 8%。每个细胞DM数目 1～ 70对不等 ,平均 5 6对。DM多出现在有染色体结构和数目畸变的细胞中 ,包括易位、断裂、缺失、双着丝粒染色体、染色体环、染色体粉碎化、内复制、多倍体、亚二倍体等。② 2 8例DM阳性患者中位生存期 (MS)为 7 0个月 ,73例DM阴性患者MS为 31 4个月 ,二者有显著性差异 (P <0 0 1 )。结论 DM出现与白血病急变、转化、预后密切相关。本研究结果表明DM表达可作为监测白血病发展。

Objective To explore the relationship between the expression of double minutes (DM) in cases with leukemia and the prognosis of this disease. Methods DM was observed in bone marrow cells after short term culture which were derived from 320 patients with verified leukemia by biopsy. Results ① The expression rates of DM were found in 8.8%(28/320) patients. Mostly, DM were observed in cell that there are many chromosome aberrations, including translocation, breakage, dicentric chromosome, ring chromosome, endoreduplication, hypodiploid, polyploid, and so on. ②During the follow up, the median survival (MS) in 28 cases with positive DM (7.0 months) were lower than in 73 cases with DM negative (31.4 months)( P <0.01). Conclusion The findings suggest that DM expression can be used as an independent prognostic marker for patients with leukemia.
