谷氨酸棒杆菌基因缺失菌株的定点构建 被引量:8

摘要 通过目标基因内部缺失片段的获得以及DNA重组交换等技术的有效运用 ,在氨基酸工业重要生产菌谷氨酸棒杆菌中 ,成功地定点构建了两个目标研究基因的单基因缺失菌株和双基因缺失菌株 ,并通过了PCR和测序等方法的分析验证。 This study is focused on Corynebacterium glutamicum, an important producer in amino acids industry,by the protocol with construction of the in frame deletion fragments of targeting genes,application of DNA recombination technique and so on.We achieved successfully the defined deletion mutants in frame of the single gene or double genes.The in frame deletion mutants are subsequently analyzed and confirmed by polymerase chain reaction and sequencing.
