摘要
为探讨蛋白激酶 C对哮喘患者 T淋巴细胞核因子 - кB(NF- кB)活化的调控作用 ,分别从 16例急性发作期哮喘患者及 16名正常对照者的外周血中分离出 T淋巴细胞并分成 3组培养 ,即空白对照组 ,加入 PKC激动剂 12 -肉豆蔻酰 - 13-乙酸佛波酯 (PMA)的 PMA组 ,同时加入 PMA和 PKC抑制剂 Ro31- 82 2 0的 PMA+Ro31- 82 2 0组。用免疫组织化学染色方法和 Western印迹检测 NF- кB和抑制蛋白 - кB(I- кB)的表达。结果显示 :哮喘 PMA组 NF- кB活化的细胞百分比显著高于空白对照组和正常 PMA组 (P<0 .0 1) ,且 I- кB水平显著低于上述 2组 (P<0 .0 1) ;而哮喘 PMA+Ro31- 82 2 0组 NF- кB活化的细胞百分比显著低于哮喘 PMA组 (P<0 .0 1) ,且 I- кB水平显著高于该组 (P<0 .0 1)。提示哮喘 T淋巴细胞 NF- кB的活化可能受 PKC调控 ,T淋巴细胞 PKC— NF-
In order to explore the role of protein kinase C (PKC) to regulate the activation of nuclear factor кB (NF кB) in T lymphocyte in asthma, T lymphocytes were isolated from peripheral blood and purified in 16 asthmatic patients and 16 normal controls, and divided into 3 groups separately: control group, PMA group stimulated with phorbol 12 myristate 13 acetate (PMA), PMA+Ro31 8220 group stimulated with PMA and Ro31 8220. The expression of NF кB and inhibitor protein кB (I кB) was detected by immunohistochemical staining and Western blot, respectively. The results demonstrated that the percentage of cells with active NF кB was significantly higher and the expression level of I кB was significantly lower in asthmatic PMA group than in asthmatic control group and normal PMA group respectively (all P <0 01). The percentage of cells with active NF кB was significantly lower and the expression of I кB was significantly higher in asthmatic PMA+Ro31 8220 group than in asthmatic PMA group (both P <0 01). It was revealed that the activation of NF кB in asthmatic T lymphocyte may be regulated by PKC, and the activation of PKC-NF кB signal pathway of T lymphocyte may play an important role in the pathogenesis of asthma.
