摘要
为探讨匹那地尔 (Pinacidil)冷超极化停搏液对大鼠缺血心肌的能量保存作用 ,并与传统高钾去极化停搏液(St.Thom as液 )进行比较 ,在改良 L angendorff离体大鼠工作心模型基础上进行低温缺血再灌注实验。把 4 0只大鼠随机分为 4组 ,每组 10只 ,S1组停搏液为 4℃ St.Thomas液 ,停搏时间为 1h;P1组为 4℃含匹那地尔的超极化停搏液 ,停搏时间为 1h;S2组停搏液同 S1组 ,停搏时间为 2 h;P2组停搏液同 P1组 ,停搏时间为 2 h。再灌注转为工作心 30 min后 ,取心肌标本置入液氮罐中 ,用荧光素酶法分别测定心肌 ATP含量。结果显示 :在保存心肌 ATP含量方面 P1组优于 S1组 (P<0 .0 5 ) ,而 P2组与 S2组相当。提示 :匹那地尔可作为心肌保护成分。
The energy preservation of hyperpolarizing cardioplegia with Pinacidil during prolonged periods of hypothermic cardioplegic arrest was studied and was compared with the traditional St. Thomas' Hospital solution. Using a modified isolated rat working heart model, 40 hearts were randomly divided into 4 groups ( n =10 each) undergoing 1 h or 2 h of global hypothermic ischemia followed by 60 min of reperfusion. Two different cardioplegic solutions were administered intermittently: S1 ( n =10), 4 ℃ St.Thomas' solution as control group, arrest time was 1 h; P1 ( n =10): 4 ℃ Krebs Henselleit solution with Pinacidil (50 μmol/L), arrest time was 1 h; S2: 4 ℃ St.Thomas' solution as control group, arrest time was 2 h; P2: 4 ℃ Krebs Henselleit solution with Pinacidil (50 μmol/L), arrest time was 2 h. ATP content of myocardium were measured. The results showed that the ATP content in the group P1 was higher than in the group S1 ( P <0 05). There was no signficant difference in ATP content between the group P2 and the group S2. It was suggested that Pinacidil might be an effective cardioplegic agent. Energy preservation of hyperpolarizing cardioplegia with Pinacidil was superior or equal to that of traditional St.Thomas' solution in myocardial protection.
