摘要
为研究血管内皮生长因子 (VEGF)过度表达在卵巢癌转移中的作用及可能机制 ,以脂质体介导 VEGF c DNA转染 2株卵巢癌细胞系 Ca OV- 3、COC1,经逆转录聚合酶链式反应 (RT- PCR)、Western blot检测转染 VEGF c DNA前后癌细胞 VEGF、明胶酶 A(MMP- 2 ) m RNA及蛋白表达水平 ;Boyden小室体外侵袭实验比较 VEGF c DNA转染 2株癌细胞前后通过人工重组基底膜的能力变化。结果显示 :VEGF c DNA转染后 2株卵巢癌细胞 VEGF、MMP- 2在 m RNA及蛋白表达水平均较转染前明显增强。Boyden小室体外侵袭实验表明转染 VEGF c DNA的 Ca OV- 3和 COC1穿过人工重组基底膜的相对百分率为 (42 .5± 4 .1) %、 (2 6 .8± 2 .4 ) % ,较对照组穿膜细胞相对百分率 (2 4 .7± 1.9) %、 (8.6±1.1) %明显增加。提示微环境中 VEGF参与了卵巢癌浸润、转移 ,其机制可能与诱使 MMP-
To study the role of vascular endothelial growth factor (VEGF) overexpression in ovarian tumor metastasis and related mechanism, the VEGF cDNA was transfected into ovarian tumor cell lines CaOV 3 and COC1. Transfected and nontransfected cells were screened for VEGF, MMP 2 mRNA and protein by RT PCR, Western blot and substrate zymography respectively. The invasive ability of two ovarian tumor cell lines were compared by Boyden chamber invasion assay before and after VEGF cDNA transfected. The expression of VEGF, MMP 2 mRNA and protein were increased after VEGF cDNA transfected, detected by RT PCR, Western blot and substrate zymography. After VEGF165 cDNA transfected, the mean invasion percentage of two ovarian tumor cells [CaOV 3: (42 5±4 1) %; COC1:( 26 8±2 4) %] was higher than that of before [CaOV 3: (24 7±1 9) %; COC1: (8 6±1 1) %]( P <0 05). It was indicated that the expression of VEGF was correlated to the in vitro invasion of ovarian tumor cell and induction of MMP 2 by VEGF may be a key factor in the process of VEGF induced ovarian tumor cell invasion.
