肝细胞生长因子治疗小鼠中毒性肝炎的实验研究

Experimental studies of human hepatocyte growth factor of fetal liver origin in the treatment of acute hepatic failure in mice

本文研究了肝细胞生长因子粗制品(hHGF-1),及经三步纯化的肝细胞生长因子(hHGF-3)对小鼠中毒性肝炎的治疗作用。两者腹腔注射使中毒性肝炎小鼠的死亡率从对照的60%分别降至11%和0,并能促进肝细胞DNA合成,使升高的血清胆红素和血清谷丙转氨酶在96hr内恢复或接近正常。组织学检查发现hHGF-1和hHGF-3有保护肝细胞、促进肝细胞再生和治疗肝损伤的作用,hHGF静脉注射对中毒性肝炎的治疗作用更明显。我们认为肝细胞生长因子是起上述治疗作用的有效成份,也是胎肝细胞输注治疗重症肝炎的主要有效物质。

A crude preparation of human hepatocyte growth factor (hHGF-1) was obtain from the subfraction of molecular weight ranging from 10,000-30,000 daltons by ultrafiltration of human fetal liver lysate. Then its further purified preparation(hHGF-3) was prepared by after three steps of biochemical purifications. The effectiveness was compared in the treatment of acute hepatic failure in mice, which was induced by treated with carbon tetrachloride, either by i.p. or i.v. injection route. Both preparations were very successful in reversing the lethality of carbon tetrachlorideinduced hepatic necrosis and reduced mortality from 60% in the control animal to 11% in hHGF-1 treated group and 0% in hHGF-3 treated mice (P<0.01). It is, therefore, concluded that hepatocyte growth factor is an effective factor playing important role for the treatment of fulminant acute hepatic failure.
