摘要
用斑点杂交半定量法对6株人、1株大鼠和5株小鼠白血病细胞以及部分正常对照细胞进行了TGF-β、TNF-α和LIF三种抑制因子mRNA表达水平的检测。得出以下结论:(1)615小鼠白血病相关抑制因子(LAI-615)属于TGF-β类因子。(2)白血病细胞与正常对照细胞相比,三种负性因子的表达存在差异。(3)LIF的表达与否,可能与白血病细胞类型有关。(4)白血病细胞中负性因子的表达强度高于正常对照细胞。(5)TGF-β较TNF-α和LIF更普遍存在并参与正常和白血病造血细胞增殖和分化的调控。
Expression of mRNA of transforming growth factor-beta(TGF-beta), tumor necrosis factor-alpha(TNF-alpha) and leukemia inhibitory factor(LIF) in G human, 1 rat and 5 murine leukemic cells and also in 5 normal cells were measured by Dot Blot sem-quantitative analysis method. The conclusions were; (1)The 615 mouse leukemia associated autoinhibitory activity(LAI-615) is belong to TGF-beta like factor. (2) Between leukemic and normal cells, the expression of three kinds of negative factors are different. (3) Whether LIF mRNA is expressed or not, it may be depends on category of leukemic cells. (4) The leukemic cells which expressed TGF-beta, TNF-alpha and/or LIF had higher level expression than normal cells. (5) The positive rate of expression of mRNA of TGF-beta in leukemic cells is the most among the three kinds of factors.
