摘要 目的 探讨基因重组hCTLA4Ig对小鼠脾淋巴细胞增殖反应的影响。方法 于不同时相点在ConA刺激的小鼠脾淋巴细胞及BALB/c与昆明种小鼠混合淋巴细胞培养中加入不同剂量hCTLA4Ig ,应用3 H TdR掺入法测定淋巴细胞增殖情况。结果 重组hCTLA4Ig能显著抑制ConA刺激的脾淋巴细胞及混合淋巴细胞反应中的细胞增殖。 结论 重组hCTLA4Ig能抑制鼠淋巴细胞的活化。 Objective To investigate the affection of recombinant hCTLA4Ig on mice splenic lymphocytes activiation.Methods Different dosages of rhCTLA4Ig was added to Con A stimulated mice splenic lymphocyte culture and mixed lymphocte reaction(MLR) system at hour 0,5 and 10.DNA synthesis of T cells was determined by 3H incorporation method.Results rhCTLA4Ig could significantly inhibit lymphocyte proliferation both in Con A stimulated splenocyte culture and MLR.Conclusion rhCTLA4Ig could suppress mice lymphocyte activation, stimulation either by allo antigens or ConA. The underlying mechanism of inhibition of ConA stimulation might indicate a novel pathway of T cell proliferation.
