摘要
建立了一种毛细管区带电泳法 ,用于同时测定5种核苷酸———鸟苷二磷酸 (GDP)、尿苷二磷酸(UDP)、腺苷二磷酸 (ADP)、胞苷二磷酸 (CDP)、次黄嘌呤核苷酸 (IDP) ;研究了缓冲液的pH值及磷酸盐的浓度对分离5种核苷酸的影响 ,5种核苷酸在75mmol/L三羟甲基氨基甲烷 (Tris) -50mmol/L磷酸二氢钠(NaH2PO4)、pH为8.00的缓冲液中可以基线分离 ;另外还研究了加入阳离子表面活性剂十六烷基三甲基溴化胺(CTAB)以及CTAB的浓度和进样时间对分离5种核苷酸的影响 ;该法成功地应用于测定核苷酸口服液中ADP、CDP、GDP。
A capillary zone electrophoresis method for the determination of five nucleosides diphosphate(ADP, CDP, GDP, UDP, IDP) was developed. The effect of buffer pH and concentration of phosphate on the separation of the five nucleosides diphosphate were studied. The five nucleosides diphosphate in the buffer solution of 75 mmol/L Tris-50 mmol/L NaH2PO4 at pH 8.0 achieved a baseline resolution. The method was applied to determine the content of ADP, CDP, GDP and UDP in the oral nucleotide liquid with satisfactory results. The effects of CTAB and the injection time on the separation of the five nucleosides diphosphate were also investigated.
