摘要
目的 探讨端粒酶、细胞周期素D1(CD1)与大肠癌的发生、进展及预后的关系。方法 收集 2 4例手术切除的大肠癌标本 ,同时取距癌灶 5cm以外的癌旁组织及 10cm以外的正常组织。检测癌组织、癌旁组织及正常组织中CD1mRNA及端粒酶的表达。结果 1.2 4例大肠癌组织端粒酶阳性率 87.5 % ;癌旁组织阳性率 4 .2 %。癌旁组织和正常组织均与癌组织有显著性差异 (P <0 .0 1)。端粒酶活性在癌灶直径 >3cm的病例中明显高于癌灶直径≤ 3cm者 (P <0 .0 5 ) ;浸润深度达浆膜层者明显高于浸润至肌层者 (P <0 .0 5 ) ;有淋巴结转移者明显高于无淋巴结转移者 (P <0 .0 5 ) ;Dukes分期高 (C、D期 )的明显高于分期较低 (A、B期 )者。 2 .CD1mRNA的表达在大肠癌、癌旁及正常组织中有明显差异。CD1mRNA的表达在浸润至浆膜层组明显高于浸润至肌层组 (P <0 .0 5 ) ;有淋巴结转移组及晚期病变 (DukesB +C +D期 )组亦明显高于无淋巴结转移及早期病变 (DukesA期 )组 (P <0 .0 5 )。 3.端粒酶阳性病例的CD1mRNA表达明显高于阴性病例。端粒酶活性与CD1表达呈正相关。结论 1.端粒酶活化在大肠癌发生过程中起重要作用 ,并与大肠癌的进展、侵袭及转移有关。 2 .CD1过度表达与大肠癌的发生、发展及转移有关。 3.
Objective To explore the association between telomerase, CyclinD1(CD1) and the occurrence, progress, prognosis of colorectal carcinoma. Methods Twenty four specimens surgically resected from patients with colorectal carcinoma were collected. Then the expression of the mRNA of CD1 and the telomerase activity in the carcinoma, pericarcinoma and normal tissue were detected by in situ hybridization. Results The positive rates of telomerase in colorectal carcinoma, pericarcinoma and normal tissue were 87.5%, 4.2% and 0 respectively, while those of CD1 mRNA were 37.5%, 4.2% and 0. There were significant differences of telomerase activity and the expression of CD1 mRNA among three groups ( P <0.05). Telomerase activity was significantly correlated with the diameter of carcinoma, invasive depth, lymph node metastasis and Dukes stage( P <0.05), but not significantly correlated with age, sex, site of carcinoma, differentiated degree and distant metastasis ( P >0.05). The expression of CD1 mRNA was significantly correlated with site of carcinoma,invasive depth, lymph node metastasis and Dukes stage( P <0.05), but not significantly correlated with age, sex, differentiated degree and distant metastasis ( P >0.05). Conclusion 1. Telomerase plays an important role in the pathogenesis of colorectal cancer, and it is associated with the invasion, development and metastasis of colorectal cancer. 2. CD1 mRNA is overexpressed in colorectal carcinoma. And its overexpression indicate a poor prognosis in colorectal carcinoma. 3. Overexpression of CD1 mRNA results in disturbence of cell cycle, then induces telomerase activation.
