摘要
目的 :观察不同剂量 X射线照射对小鼠腹腔巨噬细胞转录因子 NF- κB的影响。方法 :用凝胶电泳迁移率变化分析法得到电泳带 ,用 Phosphor Imager扫描仪分析数据。结果 :0 .0 75 GyX射线全身照射后 4h开始小鼠腹腔巨噬细胞 NF- κB的 DNA结合活性逐渐升高 ,48h时基本恢复到假照水平 ,2 .0 0 0 Gy X射线全身照射后小鼠腹腔巨噬细胞 NF- κB的 DNA结合活性从 8h开始上升 ,持续到 48h。结论 :低剂量和较高剂量 X射线均能诱导小鼠腹腔巨噬细胞中的转录因子NF-κB。
Objective: To study the effect of different doses of X ray radiation on activation of NF κB in peritoneal macrophages of mice. Methods: Electrophoresis mobility shift assay (EMSA) was used to separate the bands of NF κB protein combined with isotope labeled DNA probe, the results were analysized by PhosphorImager Scaning System. Results: The binding activities of NF κB protein to DNA began to increase graduatly 4 h after whole body radiation (WBI) with 0.075 Gy X ray, followed by return to the sham radiation level at 48 h while the increase of binding activities of NF κB protein to DNA arose at 8 h graduatly sustaining at higher level up to 48 h after the treatment in 2.000 Gy X ray.Conclusion: Both low and high dose irradiation could induce the NF κB activation in peritoneal macrophages of mice.
