摘要
目的 :构建人碱性成纤维细胞生长因子 b FGF基因真核表达载体 ,探讨 b FGF基因转移治疗视网膜病变的方法。方法 :将 b FGF和 GFP基因克隆到真核表达载体 pc DNA3中。结果 :酶切及琼脂糖电泳分析 pc FG中含有 b FGF和 GFP基因。结论
Objective: To construct eukaryotic expression plasmid of human basic fibroblast growth factor gene (hbFGF) and green fluorescent protein (GFP) for further study on bFGF gene therapy of retinitis pigmentosa. Methods: The hbFGF and GFP genes were inserted into the eukaryotic expression plasmid pcDNA3. Results: The pcFG plasmid was confirmed to containe correct nucleotide sequence for hbFGF and GFP gene by restriction endounclease digestion and agarose gel electrophoresis. Conclusion: A eukaryotic expression plasmid containing hbFGF and GFP gene is successfully constructed in the study.
