摘要
目的 :建立人工真皮成纤维细胞分离培养及鉴定方法。方法 :取出生 2 4 h内的 Wistar大白鼠背部皮肤 ,分别用组织块法及酶消化法分离纯化、传代培养 ,用免疫组织化学方法进行鉴定。结果 :用组织块法和酶消化法培养的成纤维细胞呈梭形 ,属纤维型细胞 ,免疫组织化学染色 95 %以上细胞中间丝波形蛋白呈棕色颗粒或棕色着染 (阳性 ) ;结蛋白抗体和第 因子相关抗原均无棕色颗粒或棕色着染 (阴性 ) ,表明所培养的细胞为比较纯净的成纤维细胞。结论
Objective: To explore the methods of isolation, culture and identification of fibroblasts in artificial dermis. Methods:The techniques of histotomy,degistation with enzymes and immunohistochemicstry were applied in the skin on the back of neonatal Wistar white rats. Results: With morphological analysitic methods the fusiform cells were fibroblasts and vimentin became brown granulations, immunohistochemic stain was positive and antigens found in 95% of those cells,but both antibodies against desmin eighth related with was negative. Conclusion: These techniques may provide us much purified and enough fibroblasts.
